According to the San Diego County Sheriff's Department, the accident was reported just before 6:30 p.m. on South Santa Fe Avenue near North Las Flores Drive.
Authorities said a car collided with the e-bike under unclear circumstances.
Two riders on the e-bike, including one who was described as a juvenile, suffered serious but non-life-threatening injuries.
One of the victims was airlifted to the hospital, and the other was transported by ground ambulance.
Both riders were not wearing helmets at the time of the accident.
All parties involved in the crash remained at the scene.
An investigation into the collision is ongoing.
